Been a while since I've posted here, but is sounds like you have 2 or more vertices sat on top of each other...

Band-box over 1 of the vertices, press CTRL N, then CTRL W
That will merge the selected vertices into just 1 vertex.

repeat the process for the other two vertices - then try to make your face...

Edit: (After-thought) When you press CTRL N, watch the 3D view carefully. If the shape changes dramatically, immediately undo the action and try to find the stray vertex, because if that happens, you don't have two on top of each other...

In that case then, here's a few new ideas, I'll try and work you through it step-by-step:

Idea 1:

Use the 'select tool'
Under the select options, choose faces, and make sure the 'By Vertex' box is ticked, and 'Ignore Backfaces' is not ticked
Drag a band-box over your entire model
Choose 'Hide Selection' from the edit menu (or Press CTRL H)
Any vertices still showing were not connected to faces, and can be safely deleted. Do NOT use the 'Select All' option, to do this!
Go Back to the select tool, change the selection type to Vertex
Select every vertex you see and delete it.
Choose 'Unhide All' from the edit menu (or CTRL SHIFT H)
Try again to make your face...

Idea 2:

Presumably there were no unconnected vertices?

I've attached a (slightly) annotated shot of something that should work

Select the face tool [Marked as (1) on the pic]

Single-click on each of the three vertices to form a new face between the selected vertices. After clicking the second vert, a red line will appear between the two you clicked. Do not be alarmed when you see this, it's perfectly normal. When you click the third vert, the face will be created...

(You must connect the vertices in one of the following orders:
2,3,4
3,4,2
4,2,3

Selecting the verts in any other order will create the face pointing the wrong way...)

Now you will have a new face... Try idea 1 again, and see if the stray vertex now appears - if not... it doesn't really matter, you've now created your face anyway. But it's obviously best if you can find it
