I tried to update DBP to v7.5 but I got this error. To be clear I have the full version, and it is activated. Also, DBP is on my E: drive. I tried to just install it into it's directory(Dark Basic Professional)...
Any ideas?

Try reinstalling DBPro, and then apply the update. It sounds like the registry entries were lost.
