to day in the tip of the day box on AppGameKit I saw this,

Turning off transparency for sprites that do not need it can improve performance.

how can I tell if a sprites transparency is on
and how can I turn it off?

Transparency is on by default for all sprites. I believe the command to turn it of is SetSpriteTransparency, but I could be wrong about that

its, SetSpriteTransparency( iSpriteIndex, mode )
