Firstly:
What is the name of the Windows DLL File that will allow Edit boxes buttons spinedits etc... to appear on the screen. (kernel32?)

Is there a help or reference file that will tell me how to call functions within the dll. Or could sombody list some common functions in it. e.g. making a button appear.

There are a lot of things that you cannot do directly within DBPro - message handlers are required for most GUI related things, which is why Rob wrote his BLUE GUI plug-in

Most of the required functions are in User32.dll

I'm afraid it isn't quite as simple as calling a function to make a button appear. You simply cannot do it in DarkBASIC Pro code, because DarkBASIC doesn't allow you to get function addresses (the place in memory where the function starts). Buttons etc. are all types of windows, in order to create a window, you need to fill various type structures and pass them as parameters to the CreateWindow / CreateWindowEx functions. RegisterClass is also needed. I don't think you can do this in DB either.

There are various articles on MSDN. Search for "MSDN CreateWindow" on Google for more info.
