Yes you can. But you need an additional program called a keymapper which maps joypad/gamepad keys to keyboard keys. Some joypads ship with a simple program for doing this.

But I prefer a program called Total Game Control. You can even map the analog sticks to function like a mouse (if your joypad has them).

I use an XBox 360 controller plugged into a USB port this way. I don't have a 360, I just like the controller, and it works with Windows...
