HI! How can i do a Ventilation Duct that goes along one floor
and then drops down a floor.
I tried to rotate the bend segment but that did't work.

At the moment, there is no vertical duct that I know of. (could be wrong) You may get around this by building a vertical shaft from segments and have the duct intersect it at different levels. Actually, that is quite common in larger ventilation systems. The shaft could be multiple stories high and you could use ladders/platforms/etc in the shaft to get from one duct to the other.

Just one idea. I'm sure you can think of more.
