how do to make black colour on Sprite to be transparent?

your sprites should automaticaly turn black invisible
if not :

set image colorkey 0,0,0

be sure that your sprite is actually black as in rgb(0,0,0) black!!!

My advice would be - don't use black for transparent, use transparent for transparent! Load it into a decent art package (Paint Shop Pro, Photoshop, etc) and use transparency to achieve the result you want.
